In my personal experience, dissociation constants of fluorescently tagged oligodeoxynucleotides can be measured quite easily using fluorescence methods by fluorescence intensity or fluorescence polarization. Given a spectrofluorometer with a temperature-controlled cuvette holder, melting temperature can be measured the same way. You have to remember that the fluorescent tag could have an influence on the association, however.

The historical method for measuring DNA hybridization/melting is by hypochromicity, which is a decrease in UV absorbance in double-stranded DNA versus single-stranded DNA. A spectrophotometer with temperature control is required for melting measurements.
