I am about to attempt to get genomic DNA (for genotyping by sequencing) from frogs that were fixed in formalin and subsequently stored in EtOH, and I was wondering if anyone had recommendations on the best tissue type to use. Do some tissues retain DNA during formalin fixing better than others?
I am using the Qiagen generead kit, btw.

Hi there Donald,
I think it's going to be difficult to isolate good quality DNA from formalin-fixed tissues. I've attached a great paper that talks about all the nasty things formalin does to nucleic acids. It sounds like even if you're able to get DNA out of the sample, it's not likely that it will amplify/sequence/etc.
