Yes, what matters is the type of lipase and substrate you are using in lipase assay. Lipases act optimally at alkaline pH say 8.5, use 0.05 M Tris buffer and a long chain fatty acid substrate bound to 4-methyl umbiliferone for flourimetry or p-nitrophenyl group for colorimetric assay.
