I want to amplify the matK region (together with nrITS) for phylogenetic species delimitation from dry-leaves of wild populations of Vaccinium floribundum.

I have been checking literature for that, but it looks like there is not an agreement in the primers used in different articles, so I am not really sure. Tsutsumi (2011) used trnK-3914F and trnK-2KR, and internal primers for sequencing. This is based on a set of primers proposed in this part of the chloroplastidial genome from Johnson & Soltis (1994).

So I was kind of confused and not sure what to order. Before I design my own primers, I wanted to know if, according with your experience, you know about specific primers for this species (or genus) that are suitable for the amplification of that gene.

Thank you,

Johnny
