We're trying to decide between the QIAamp Viral RNA Mini Kit, the standard RNeasy Kit, and just using Tri Reagent, which I believe is not a kit. Does anyone have any suggestions or other kits/methods they prefer?
Additionally, we're thinking about doing both the supernatant as well as cell lysate for viral RNA, but would the above methods work for both supernatant and cell lysate?
Zymo Research have an excellent kit for isolation of viral RNA. We have used it successfully to extract Pegi Virus from plasma samples
Tri reagent or Trizol uses a single step chemical phase-extraction for RNA prep and it has worked very well for robust large preps. If done right you get quite a lot of material out, but the draw back is that it might not be very "clean" for down-stream uses where material of a high purity is needed. We have in the past successfully combined Tri reagent, followed by a column-based clean-up to get RNA of higher purity. For that the standard RNeasy kit from Qiagen worked just as well.
