I'm trying free-floating IHC instead of slide-mounted for mouse brain sections and am currently trying to figure out the best blocking media for my purpose. Beyond something like an appropriate serum based on secondary AB, is there any reasoning behind certain blocks being better for certain stains, or is it all just trial and error? I have enough samples for trying a few different things, but wasn't sure if there was a more logical way to approach it. I know it varies for different ABs, but figure if there's a better way to start I can save time and reagents. Thanks!
