In Z-scan technique, we often use motor (digital mcrometer transformer) to move sample. I think that motor is ued to automate measurement. If there is no motor, for example, we can move sample manualy on bar (marked by lines seperating 1 mm, sample is placed on holder, holder can move on bar), we determine position of sample by reading lines on bar, and get laser intensity on detector.
So is it necessary to use motor ? Can we move sample by hand ?

It is always not necessary to use motor automatically. The idea of using this method is to save the time and energy. I have experienced by performing this experiment by stepping the motor manually and it was really big headache. Say, if you want to take some 100 readings in the open or closed aperture, you've to move it for every single data and note it down. But, now I have switched it for automatically and it seems very easy to perform the experiment. However, there won't be any difference in the results. We worked on both the methods and you can refer our articles. Good luck.

You are right, it is not necessary to automate the z-axis scan when imaging any sample. However, manually altering the z-axis by hand obviously takes longer to execute and will mean you take longer to image each sample. Additionally, you run the risk of introducing a random error into your measurement by not moving the z-axis precisely 1mm each time. This may lead to more problems later on when it comes to analysis as you will have to include larger errors into your final results. With an automated method of moving the z-axis, there may be an error in moving 1mm each time but it will be the same for all samples and measurements meaning that it is systematic rather than random. It is therefore easier to incorporate this error into your final measurements and usually leaves you with a smaller final uncertainty.
Automated methods are fairly simple to implement (and can be run separately to the rest of your software if required). They can also be very inexpensive. For something as coarse grained as moving 1mm, a stepper motor attached to your z-axis screw would suffice.

You can use both ways. Automatic stage as well as you can move sample by hand. When i did Z-scan experiment i used both ways, i did not find any difference, however the automated stage will give the precise readings of the sample positions.

it dependes on how often you will do the scan.
Motorizing a scan is in general a time consuming task, even when controlled by a computer.
So it may be often simpler to do it by hand.

Using motor only increases the rate of measurement. I do Z-scan measurements by hand. That is possible.

If you use a stepper motor you can increase the speed of scan and thus you can eliminate any possible thermal response that appears by overexposure of the sample to the spot laser.
