If you are interested in the surface details of the cell wall, you'll likely need SEM. If you are interested in its cross-sectional details, you'll likely need TEM.
I am currently working with enterococci and lactobacilli, I am looking on the one hand to observe their cell form before and after stress and on the other hand to observe the cell wall before and after the same stress, what should I use? if I use the SEM can I use a liquid sample for the metallization or do I have to have a lyophilisate? At what magnification for each observation (shape / wall) Please!!
