I have isolated aspergillus niger from cow dung. it shows negative results for amylase, pectinase etc. however In literature I studied positive results for this strain. is this possible or am i not right about the screening tests?
Although it is an Aspergillus Niger strain, the performance of producing enzymes varies greatly among the strains. It is a normal matter that the strain you screened does not produce amylase and pectinase. So, It's okay! please further screen other strains that can produce amylase and pectinase.
It's quite likely that your A. niger fungal strain does not produce any of these enzymes. However, if you subculture the fungal strain on casein, starch or pectin cotaining plates, the strain may show some of these activities.
Are you sure that the mold is even Aspergillus niger? How did you arrive at the species name? It is a rare/strange occurrence that a strain of this mold does not produce any of these three enzymes. What screening methods did you use? Please perform the screening experiment again after confirming your mold is Aspergillus niger.
