Hello, We have had a contamination problem with our primary cells. The media remains red, which is making me think that we could exclude bacteria. It takes at least one weak for that contamination to appear. It creates a white powder. Contamination are isolated round spheres. Also, our other cell lines which are not in this incubator for primary cells are perfectly fine and I have never seen this contamination with those. I am thinking it might be a problem with the incubator for primary cells. Does anyone knows how to really clean an incubator? Also has anyone ever had this kind of contamination

How big are the spheres? It might be yeast. At first, I would recommend checking the cultivation media and other chemicals. It is of course important to keep incubator clean, bud as a source of contamination I would say it is one of the latest options. Is this contamination occuring regulary? If it is single case, I would say it was just bad luck, there is always a chance that contamination will appear during your work

As the friend Jakub said, is like me contamination by fungus. We had a similar problem some time ago, but in our case we decided to discard the cultures and all mediums were being used. Regarding the incubator cleaning, clean all shelves with soap and chlore, and posteriorly, with alcohol 70% v/v. Then check the incubator's filter. Change it regularly.
Things that are round and look like little grapes are generally yeast, do you use a anti-mycotic in your media? I usually use a antibiotic/antimycotic combination for my media. Works very well, you can buy the mix commercially and just add to media when you make it up.
When nothing goes right, just discard everything. clean up and start fresh. It is never too late to start again properly.
