I stained cavernous nerve with toluidine blue, but the samples were violet like if i had used cresyl violet coloration. so, i would like to know if can use toluidine blue to observe neurones regeneration or degeneration in brain?
While toloudine blue is a technique that can be used, we recommend using Cresyl violet, which stains better reticulum and the nucleus, as toludine blue behaves differently depended the chemical nature of the substance to be stain (orthochromatic stain (blue) or metachromatic (purple-red)
Toluidine blue is more like blue than Cresyl Violet. Cresyl Violet has purplish blue red coloration. You can also try Methylene or Methyl Blue which stains neurons very nicely. Good luck.
thank you two for your answers. But Ranu, can I use methyl blue instead of toluidine blue.In this cas, should I continue with the same staining procedure or there is a specific procedure for methyl blue? thank you
