I have revived the MCF10A cells after a long time. They seemed fine after revival but after the first passage, I find these black dots floating in the suspension. Earlier I read that over trypsinization may cause the appearance of the black floaters due to cell debris. But this was not in my case. Moreover the cells don't seem healthy to me. I also observe few vacuoles in some of them.
Anyone working with MCF10A may kindly comment on the cells please.
You can count the cell viability using trypan blue exclusion. Dead cells take up the dye within a second making them easily distinguishable under the microscope from viable cells which remain unstained.
Thank you Moslim Mohsin Khalaf . I have done that. It is fine that way. But I was concerned about the morphology and the black floating particles. Any idea about them?

I think it's debris of dead cell that affected by freezing and throwing.
