I am working on BV2 microglia cell lines. For my work, I will be stimulating the microglia with ATP on a range of concentrations. Thus far, the ATP does activate microglia as proven by Griess assay, however I would like to know whether there are any co-stimulants that can be used to improve the stimulation of BV2 microglia.
Hi, I am also working on the BV2 cells. May i ask you how you prepared the ATP solution.I purchased ATP powder from Sigma. But BV2 dead quickly after ATP treatment. I am wondering do i need purchase ATP solution from Thermo directly?Thanks.
Hi, I have prepared the ATP as instructed in the data sheet. and yes, I have purchased mine from Sigma as well. the death of cells might be due to the high concentration of ATP used. what is your concentration of interest? I have used mine ranging from 50, 100, 250, 500, 1000, 5000 uM. only at the highest concentration, 70% cell death was observed.
Hello,I treated LPS 1ug/ml for 4.5h and ATP 5mM for 1h in BV2, NLRP3 IL-1B can be detected quite well in PCR and WB.
But the cell death measured by LDH release and PI uptake were not obvious.Iam confused too
