are LB broth and Nutrient broth minimal or complex media? I am following the QIAGEN protocol for the RNA extraction from bacteria and the protocol for the preparation of lysates is different depending on whether the medium is minimal or complex . I have to do it on Klebsiella pneumoniae , that is a Gram negative, and I usually grow this bacterium in Nutrient broth overnight.

composition of both medium is almost the same (minimal medium) . optimal culture medium for K.pneumonia is NB .

Hi Silvia, by basic definition, minimal medium is that which has just basic needs for the growth of bacteria like inorganic salts, carbon and nitrogen sources along with water. These are the core nutrients essential for growth. In this regard, if you simply check the compositions of Nutrient and LB broths, they are not fitting at all to the definition of minimal media as both have complex ingredients in their compositions like peptone, tryptone, yeast extract etc. So they are basically classified as complex media but both are non-selective as well, as numerous types of bacteria can grow in them and you cannot differentiate. So here for your target Klebsiella its better to choose a complex media not the minimal one from the point of view of easy growth in lab. But anyhow if you want minimal media so Simmons or Koser Citrate Agars or broths are excellent to work with, as they are used widely for the differentiation of Enterobacteriaceae members and Klebsiella can grow substantially on it by utilizing the Sodium Citrate of the medium as sole source for carbon and Ammonium Phosphate compounds as sole nitrogen source, which can be visualized by blue color from the original green color of the medium and this is due to increase in pH of the medium after the bacterial growth due to excess carbonates and bicarbonates. But on minimal media incubation time often matters, it may be from 24 hours till 48 hours for the proper growth to be seen.
