I use LS 180 cell line that already trypsinized. I read in ATCC, LS 180 have never been trypsinized. so after that, I never use trypsin but my cells easily detach now. What should I do?
I use trypsinization for both primary and stable cell lines. These days I am working with HEK-293 cells and have faced a problem like this. There are few things that might be helpful, the first one is to coat your plates with Gelatin before seeding the cells and other is to handle the cells very gently while lifting up the plates. If you are totally new to this cell line try to give them 2 to 3 passages before you start your further work. Sometimes they take time to get used to media and conditions once started with frozen stocks.
