I am trying to detect tau in lysates of SH-SY5Y neuroblastoma cells using this antibody. However, I am having trouble detecting any bands. Once or twice I got faint bands, and most of the time I do not get any bands at all. Anyone has had any success using this antibody?
I am loading about 100 ug total protein on the gel. I have tried antibody dilutions ranging from 1:1000 to 1:500. On the antibody vial, they have not given the concentration, only the total amount (100ug) and not mentioned the volume. So I am unable to estimate the concentration of the antibody stock.
I've used a rabbit monoclonal from Abcam to detect tau. I don't recall the exact clone name or concentration. The bands were very clear and I was also working with SH-sy5y cells. Hope that helps a little
Thank you for your input! May I ask what buffer you used for lysis? I have tried both Triton-X based buffers and RIPA.
Hello Anuradha Venkatramani , I am also working with Tau 5 antibody to detect total tau from SH SY5Y cells. I have no luck detecting any tau. Did you figure out why it was not working for you? Could you help with some insights?
