I am using negative sorting to isolate astrocyte but having some problem and i was thinking should i go to positive cell sorting?
Does positive cell sorting has any superiority over negative cell sorting technique?

This is not my expertise, but positive selection Means that you will use antibodies against surface markers that are present on the cells of interest (Those you want to isolate), Whereas negative selection Means that you will use antibodies to bind cells you want to get rid of and you will obtain your cells of interest because they will be the only ones unmarked, not fluorescent. Sometimes during positive selection the cells of interest become activated by the antibody andyou do not want that! the Ab
I want to add that you should reason what are you doing with your cells afterwards, to decide if you can change your strategy from negative to positive selection...

I also thought that positive selection might make an cell activated.
as iam working on astrocyte and believe that it gets activated by positive selection but i donot have any paper or anything to take it as true.
i will be using astrocyte for single cell rnaseq and believe that neg sorting might help in maintaining the cell integrity for the seq experiment.
