Specifically looking to replace the clickit kit with homemade buffers- almost out of buffer 1 but still have plenty of the rest for now. Does anybody use a homemade click system and willing to share protocols? Thanks
I'm not familiar with these kits, but worked with click reaction before. After looking up on this protocol I assume that buffer 1 is an accelerating ligand. We used PPh3, but for biological samples this paper mention THPTA. I think it should work more or less with any other accelerating ligand.Article Copper-Catalyzed Azide-Alkyne Click Chemistry for Bioconjugation
For general click reaction you can download a protocol here https://clickchemistrytools.com/wp-content/uploads/2016/04/Click-Chemistry-Protocols_rev2.pdf
There are low cost alternatives (the same click-it kits under different name) to some Click-it kits, you can browse here https://clickchemistrytools.com/product-category/click-chemistry-toolbox/click-chemistry-toolbox-auxiliary-reagents/
