I'd like to quantify total TRIF protein in my murine macrophages. My previous experiments using an anti-TRIF polyclonal ab in Western blotting have given me unclear data because I have multiple bands and do not have a trustworthy positive or negative control. Does anyone know of a way to reduce expression of TRIF in macrophages, or of a cell-type that has expresses a high level of TRIF? Also, does anyone have a reference for an excellent anti-TRIF antibody? If not, I am going to have to try siRNA....
Clone 1H4B01, isotype mouse IgG2a, k. I have been able to detect TRIF in HEK cells and murine BMDMs. The band is at 100kDa instead of the uniprot predicted 76kDa. Results were confirmed in flagged-TRIF hek cells and by using small molecules to reduce TRIF total protein. The reference is biolegend catalog 657102, purified anti-ticam-1 (TRIF) antibody.
