I am having issues with RIN values I am getting with my RNA samples I extracted from postmortem brains. I got RIN values on 2 machines and the results were somewhat close: all above 7 and within 0.5 - 1 points between replicates. Now I sent my samples outside to a company for sequencing, I figured since all my values are >7 I would save some money and go for polyA pulldown. Now the company did QC and Bioanalyzer and got RIN values that are all at least one point below my original reads. Some are more than a couple of points off. But all the DV200 values are between 88-92%. Unless a -80 disaster happened that I dont know off - which is unlikely- what might explain this discrepancy? 5 of my samples have RIN of 4.8-5.8 which is way below the accepted threshold. Any thoughts?
