Are there any other methods besides ELISA for Interleukin-7 detection and quantification? Can we quantify it through HPLC?
Hi Bapi You can also try one of the gene array or multiplex systems. This will screen a whole panel of cytokines simultaneously. However it is very expensive. Flow cytometry is also used for this. Would stay away from Western Blotting as this system is not sensitive enough to monitor culture supernatants.
Thank u Mr. Pool for your suggestions. We are currently using the ELISA Kit for quantification and detection. However, I am interested in knowing whether the same can be performed using RP-HPLC.

If you have in hands a strictly IL7-dependent cell line together with an IL7 standard, proliferation assay might be used for quantification.

Actually I work on yeast system for the protein production. Though I am using ELISA for its detection and quantification, I am also looking for the alternatives. I have come across a paper stating the RP-HPLC technique for the same. However, the information was scarce.
I would like to know the aforementioned (RP-HPLC) technique in details.

So you have every details required to set up the experiment. All you need is an IL7 standard to calibrate he detection system (to make the link between the area of the peak detected an the actual quantity of IL7 injected).
