There exists a dual growth characteristics for Rhodococcus opcaus PD630. Though at suspended growth, the OD is read easily I am finding rather difficult in reading OD during aggregated growth. Kindly help
Can you do dry or wet weight measurements? Even if it were not difficult to measure OD during aggregated growth, the change in cell shape would change the relationship between OD and cell density and make it difficult to draw any conclusions from the data.
It is always better to measure the OD of a microbial sample when it is homogeneous, in order to establish the relationship between the OD and dry biomass weight properly. I suggest that you can try breaking the aggregate by providing magnetic stirring for like 1-1.5 hrs. You should keep the stirring at r.p.m. 50-60 (not greater than 80), as it may damage the cells due to shear stress, if higher r.p.m is used.
