I have used two different BDNF antibodies in the past with mixed results:
- AB1779SP (Millipore; 1:500) works OK for WB detecting the monomer at 18KDa and homodimer at 27-28KDa, with some additional unspecific bands (you may want to use a higher dilution). In IHC seems to work fine in fixed sections from hippocampal slices, although I only tried once.
- sc-546 (Santa Cruz; 1:200) works well on WB but haven't used it for IHC.
You can check Fig. 2E in this paper to have an idea how it looks on WB: http://jcb.rupress.org/content/210/7/1225.long
If you do not have luck with the commercial antibodies, Amgen has an antibody that works beautifully at very low concentrations (rabbit anti-BDNF
antibody @ 1:6000 ). Sometimes they will donate antibodies for research (particularly in the case when the commercial antibodies are not as effective).
