We isolated 3 bacterial cultures from a soil sample and kept it for storage in refrigerator for 5 days. At the end of the day, two plates were totally in a molten stage. Can anyone tell the reason for this to avoid such incidence in future? The possibility of a power cut or improper handling is unlikely.
Any possibility that the bacteria produced something to change the pH? We were trying to make agar at a variety of pH levels and the lower pH would not solidify (I think lower than 4).
But mam it was kept in refrigerator ,so most probably bacteria would be in inert stage ??
If the agar plate used and melted , this may be interpreted as hydrolyzing enzyme production that may cause plate hydrolysis and be melted
I think composition of media must be known to give any suggestion or comment in this regard. Can you pls let us know about it? or what type of bacteria you are dealing with?
