I'd like to use synthetic RNA spike-in to check for (micro)RNA input and RT efficiency to minimize quantification error. I'm analyzing microRNA using qPCR. Any experiences (kits) and recommendations (ID, manufacturer) are welcome.
qPCR means stem-loop primer TaqMan RT-PCR? Because qPCR is a very general term and does not explain what you actually measure.
Besides that I can recommend the Applied Biosystems miRNA TaqMan assays, they have the synthesized mature miRNA available too (call them pre-miRNA for some odd reason though).
Yes, i'm using stem-loop primer TaqMan RT-PCR! It seems that applied biosystems is offering this kind of synthetic microNA as microRNA "mimics" as well! Thanks for this hint! But i couldn't find any manual which describes the quality control in detail - any further suggestions!?
