I have been thinking of a construct, which is single-inner membrane-spanning protein, the N-terminal domain of which I want to get rid off after enzymatic activity I will induce in the E.coli cell. But still I need the full-length version of this protein to retain in the inner-membrane.

From the literature, I know that I can use signals such as PelB, OmpA signal peptides or inner-membrane-leaky E. coli mutants that will do that job, but still it is worth asking I guess.
