Does anyone have good reference papers looking at the increase in leucine concentration after a standard meal (leucine gavage) in humans and mice?
Into food, leucine is often present in small amounts and digestion of proteins, so absorption, is slow and partial, and you may not find significant plasma modifications. And, keep in mind the dye effect of small amounts of absorbed leucine in a large volume (41-42 mL of plasma for any Kg of BW), that have to be summed to significant basal values and to hormonal dependent altered metabolism of amino acids linked to the many food components. So, in my advice, if you want to increase p-leucine significantly, you have to provide it in free form. By using different amounts of leucine, you can even plan the dimension of the pattern and the length of modifications, that are,obviously, more precocious and evident with increasing levels. I suggest you to start with a minimla dose of 3g ( and 300 mL of water, in fasting individuals) for an average 70 kg human, to observe a short term (20minutes?) but significant pattern modifications. But, with growing loads, modifications can be observed as soon as 5 minutes post ingestion, and significantly positive pattern can be prolonged as much as 240 minutes.
