I am using CHO cell to express protiens, I used GMEM media (no glutamine) with supplements
I need the exact protocols to follow for suspension and adhertent cells.

If you culture in shake flask 125, Then working volume should be 30 mL, Inoculation cell density 3 to 5 x10^5 cells/mL, 8mM GlutamaX (From 200 mM Stock i.e. 1.2 mL in 30 mL), 1% Anticlumping agent (i.e. 300 uL in 30 mL). Passaging when cell density reaches between 20-30 x10^5 Cells/mL (This will be reached between 3-4 days). After 7 passages prepare stocks.
