Allah Nawaz Unless you are doing aldehyde-fixed cryostat frozen tissue sections, you DO NOT need antigen retrieval for frozen sections.
Hi,
I can answer with my experience in mouse embryo.
Some epitopes require Ag retrieval step (anything nuclear most of the time).
To do so, I use pH 6 or 9 solution depending on what works best with the specific Ag.
However be careful if you are using multiple Ab for the same experiment, some epitopes do not support Ag retrieval and you will lose signal for them.
Amplification will not be necessary unless rare cases. If so, I use a TSA kit.
