Hello, I am about to titer an antibody (and I have little experience with this). If I received a stock concentration of 2 mg/mL and the recommended dilution for my work is 1:200.....is this 1:200 of the stock solution? I don't understand how to convert the 2 mg/mL to a ratio (or if I even need to). Help??
Yes, this is 1/200 of your stock.
1/200 means 1 part of your antibody stock in 200 parts of total volume. The same can be said as 1 part of your antibody stock + 199 parts of your diluent.
For a practical example, if you need 1 ml of your antibody solution at the end, you should mix 5 microliters of your antibody stock with 995 microliters of the diluent.
The recommended dilution is a different one for each antibody stock. If you change your stock, the recommended dilution would be a different one.
Thank you, Gertrudis! I just wasn't sure if I needed to do any other calculations. Until now, I've been working with antibodies in which the recommended dilution is given as µm/mL, not as a 1:whatever dilution. So, when I have the stock concentration given as mg/mL I just use the C1V1=C2V2 calculation to convert. This is even easier!
