Hi Asif, If you can endogenously label your target protein with a florescent tag, the recovery of flourescence after inhibition can be assayed. However, the period of inhibition should be greater than the half life of your target protein.

Alternatievly, SNAP-tag and HaloTag based protocols are available as a non toxic alternatives to the orginal cyclohexamide chase assays.

Currently the best alternative to pulse chase labeling is Click-chemistry. You can do exactly similar experiments with this technique as classical pulse-chase but instead of using radio-labeled amino acids this techniques used chemically modified amino acids which can be detected on a western or gels by simple click-reactions.
