I am wondering how long does it take for baked tissue slide to start oxidizing after baking? How long can I leave them at room temperature to cool before storing them at 4C without significant oxidation?
As a rule of thumb precut slides should be stained after a minimal time of a week, when stored at room-temperature. I recommend to omit the baking after cutting and rather bake them shortly before deparaffination. Only be sure, that there is no residual water under the section. Store them vertically and clean.
The advantages are, that the tissue is still covered with paraffin while storage, the antigen is not influenced by higher temperature and uneven durations of heat and that the paraffin is melt just before deparaffination for a better result. So the slides are treated in a "more" similar way before staining, regardless of the time of cutting.
I think one can put the slides in the refridgerator just when they are dry.
