I am trying to find a very specific co-localization of clathrin on hela cell plasma membranes using the X22 mouse anti-clathrin antibody, but have so far been unable to do so. I have tried using 2% formaldehyde/0.2% triton as well as methanol fixation procedures. My adviser has suggested an acetone permeablization. Is acetone good for looking for co-localization of clathrin and other proteins on the plasma membrane?
