Issue:
According to the datasheet provided by sigma this compound gets dissolved in Methanol and Glacial Acetic acid.
I did try using both and with methanol when i made 100mM (176mg for 10ml) in methanol it did dissolve.
But when I use any lab wares made of plastic(Tips/96 well plates/Falcons) to transfer aliquots of this methanol dissolved 4-MU I am observing white precipitate formation. (All the stocks and workings are freshely prepared).
When I tried dissolving it in glacial acetic acid (17.6mg in 1 ml), it is not dissolving rather forming white crystals.
Kindly share your experience using 4-MethylUmbelliferone.

Goutham: I suppose you are using 4MU as a standard in measuring enzymatic activities that hydrolyze 4MU-substrates. For that purpose, you only need a very diluted stock solution of no more than 1.0 mM (1.76 mg in 10 ml) in DMSO. We made this stock, aliquoted into 1 ml in 1.5 ml plastic tubes and stored in 4C for years in a brown bottle preventing light. When in use, we made a "sub-stock" of 100 x dilution in water (10 microM) which can be stored at 4C for a month or so. In actual assay, we then dilute 10x with a stop solution (pH 11, final concentration = 1.0 microM) for the enzyme assays and read in a microfluorometer. It's that simple.
