10% Neutral Buffered Formalin on bacterial cells and its effects on Propidium Iodide?

I would like to know if there are any detrimental effects of fixing bacterial cells in 10% Neutral Buffered Formalin (NBF) prior to staining with Propidium Iodide (PI)?

Will this give a false positive signal of 'dead' bacterial cells.

If fixation is a requirement, should I stain with PI first and then fix?

If you are using the PI for flow cytometry to distinguish the live and dead bacterial cells.you should not fixed the bacterial cells with 10% Neutral Buffered Formalin prior the staining procedure. Formalin will fixed cells and stop the live activities of the cells.
