Hi all, I would like to ask if anyone has experience with the brain tissue that was infected with Zika virus?
We are currently thinking about performing the in situ sequencing technology on such samples (fresh frozen mouse brain), however, we do not have experience with such samples. We are wondering if the fixation with 3.7% PFA for 5 min (potentially the Ethanol series) would destroy the virus or it still could be serious? (not only dangerous for people to get infected but also to know which level of biosafety for these samples we need) Thank you for all your answers.

We used to do same experiments in my lab, but with slices of mice brain. After removing the medium, we let the PFA 4% for one hour and then rinced with PBS. After this kind of treatment Zika virus was no more infectious.
