I am going to isolate Campylobacter from cloacal swab of broiler. My queries:
- Will Campylobacter spp. grow in Nutrient Broth without any supplement?
- Is it required to use an incubated broth culture for plating with antibiotics disc in Muller Hinton Agar plate? Or I can use an incubated colony homogenised with distilled water immediately after isolation from a specific agar plate?
- Is it possible to perform antibiotic sensitivity test by Disc Diffusion method in Muller-Hinton agar without any supplement such 5% sheep blood?

Interesting work. I have always seen colleagues grow campy on Karmali Agar with low oxygen and at 42 Celsius. Campy is always a bit complicated to grow.
As for antibiotic testing, I am not sure how it works for campy! all the best.
Dear Mehedi, I suggest you to contact with Dr. Binaya Bhusan Nayak Pricipal scientiest and HOD or Dr. Sanath Kumar Senior scientist,or Dr. Manjusha Lekshmi Scientist of FRHPHM devision at ICAR- Central Institute of Fisheries Education or Dr. Srinu Rathlavath they are the expert on working with this area.

My view is that Campylobacter ssp. are tricky to isolate but doable.
1). Creating and maintaining microerophilic conditions is very key. Can use Anoxomat or in resource scarce setting a candle in the anaerobic jar
2) The broth used in enrichment largely depend on the source and background of the sample, bolton Vs Preston (new ISO methods are clearer)
3) Growth media. mCCD is mainly used but the shot fall is that all the colonies including the interfering microflora produce white colonies. It may not be easy to isolate camplylobacter colonies but I always used another very selective media; Rapid campylobacter agar that produce brick red colonies.
