as I see this question I guess it could be a little bit difficult to answer your question for several reasons (regarding basic blue 141 etc.). I have found the old paper of Kass 1987 (Am J Clin Pathol. 1987 Oct;88(4):436-41.), a Journal which offers downloadable pdf's only from 2000 on. So -if you don't have the article you should try to retrieve it from a library.
Unfortunately I did not find any reference/article using this combination of dye-cells, no mention of dye basic blue 141 in CONN's Biol.Stains,10th Ed (Chapter HOROBIN, Oxazines and related dyes), but found a recipe without a reference (Naphthol AS-D Chloroacetate Esterase method for neurophils) and did find a link to a US-patent (KASS L: Oxazine stained lymphocytes and method)
Thank you Wolfgang Muss. I think , Human alpha defensin-1 antibody staining will be my ultimate option for neutrophil granules staining. Once again thank you for your generosity for sharing this information with me.
I wrote my answer really "last minute" before leaving the lab, and since you referred primarily to a "classical histochemical stain" I did not include options for IHC...
Hope you'll be successful, would be nice to read sometimes how your results with a-defensin-1 Ab were...
