I would like to perform a transwell migration assay on breast cancer cells (4T1/410.4/NMuMG) using different pore diameters. Would anyone know if they are too large to pass through a 3um pore? I know 8um is the standard.
The answer is yes, but it depends on the assay and only under certain circumstances. My group, for example, can't get breast cancer cell line to pass through 7 um pore filters in the presence on chemoattractants or flow pressure (http://www.ncbi.nlm.nih.gov/pubmed/25614802). However, in vivo or in vitro, the inclusion of pressure forces and attractants can induce tagged breast cell lines to passing through 3 um capillaries/pores (Yamauchi, et al. Can. Res. 2005 and Sharma, et al. Intravital 2012). I would try 3-8 um and see what you get on your specific assay.
