Hi, Does there has anyone know the working PH range of the lyso-tracker probe?
I want to distinguish secretory granules and lysosomes for analyzing their interactions, but I just realized that the lyso-tracker probe could only label acidic organelles, which meant it may not have the ability to distinguish granules and lysosomes. But the PH range of secretory granules and lysosomes may be different, therefore, I want to ask for help.
If anyone know about this, or have advice, I would be so appreciated for your kindness!

So LysoTracker will accumulate in any cellular environment with lower than physiological pH. As the fluorophore is linked to a weak base permeability most likely changes based on protonation once inside any slightly acidic organelle. However, this also implies that the result of the staining is very dependent on the amount of LysoTracker used, preferentially low nM range, and the diversity of acidic organelles present in your model.
If you assume difference in pH of your granules compared to lysosomes I would suggest you employ Lyso sensor, which have reported pKa values and change emission spectra based on the pH.
This might help you to track and distinguish your granules from Lysosomes.
