I am making retrovirus with pBABE-GFP and ECO packaging vectors in the clonetech packaging cell line GP2-293. I noticed that my host cells are not green. Is this ok?
What do you mean by "be green"? They should exhibit green fluorescence when excited by blue or UV light. If you can't detect pBABE-GFP expression by fluorescence microscopy, something is certainly wrong.
If you mean, should the cells look green by eye, they can, but mileage can vary. Not in a cultured mono-layer. It's too thin, and phenol red in the medium gets in the way.
If you detach them and spin them down, cell pellets always appear yellowish-green. When you express GFP, you can often see an extra-green hue on top of that, but you need a trained eye, or better a control, to be sure. So by eye I wouldn't worry about it.
