I ran 2 cell proliferation assay on cardiac fibroblasts under varying treatment conditions (cortisol 0.5uM, Cortisol1uM) (TGF-b1, TGFb1 + Cortisol 1uM, TGF-b1 + SB431542) the results suggest that the majority of these treatments decreased metabolic activity/ proliferation yet when I analysed the same cells under same conditions on a LDH-Glo (promega) assay it demonstrated a decreae in LDH release in all treatment groups.

If cells reduce proliferation, then they also decrease metabolism. This means also that they decrease the uptake of substances.

I interpreted your results such that the treatments decreased cell proliferation and metabolism, but did not detectably disrupt the plasma membrane leading to cytosolic LDH released into to the culture medium.
