Hello everyone, I am treating U937 cells with PMA at 100nM concentration for 24 hr and almost 90% of cells get attached. Then I am culturing these cells with 10% FBS media for the next 24 hr. During this time and further, these cells keep on detaching and only ~20% cells remain attached after 24 hr of LPS or IL4 treatment. Does anyone have experience this problem? Please suggest some solutions. Thanks in advance.
I don't have any experience with this specific cell model, but withdrawal of PMA can cause apoptosis in some models, so it's probably dependent on PMA dosage and length of differentiation, but also subsequent LPS/IL-4 treatments can be a cause.

Dear Sumet, were you able to solve this problem ? I have now started using U937 cells and i have attempted differentiation with multiple PMA doses, but they always attach very loosely and the cells detach almost completely when I change their medium
