Hi Subhash: I assume you are referring to the Cre-ERT2 allele. This fusion contains a (mutant) fragment of the estrogen receptor that can bind tamoxifen but not estrogen. I this way, Cre can be activated in vivo or in vitro by Tamoxifen (eg. by injecting tamoxifen or by gavage) and Cre will be activated but it won't be activated by endogenous estrogen. Check with others but this is my impression.

That sounds very logical answer to me. Otherwise estrogen will binding each cell.
