Here's my recipe for enzyme assay (10X):

1 M BisTris

0.5 M CaCl2

At first, the pH value is 4.8, after 10-fold dilution it became 6.04, though it almost reached my desired point (pH 6). For further confirmation, i also used universal indicator sheets and found that they situated at 4-5 and 6.

As I know that the calcium ion is not as simple as alkali metal ions (IA group), and dense salt concentration will affect the detection of pH via potential difference, are there any principles/cautions for IIA metals during preparing such solutions?
