Hello researchers
all of us know, that we need a special protocol to extract DNA from frozen blood other than protocol which used to extract DNA from fresh blood .
the question is : .what are the specific changes in blood properties that obligate us to use a special protocol to extract its DNA.

DNA extraction requires separation of the white blood cells. Freezing, preservation and thawing lead to hemolysis of the red blood cells and release high amount of hemoglobin. The proteinase K activity is wasted on hemoglobin degradation when DNA histones and coagulation factors not damaged.

DNA extraction is targeted to WBCs. Freezing and thawing of the sample of frozen blood lead to destruction of RBCs. This will lead to accumulation of hemoglobin which interact with proteinase K enzyme activity. Thus we need to specific protocol avoiding the above mentioned event. Hope to be benefit answer
