I have done DNA isolation of sugarcane every time I don't get any bands of DNA can anyone tell me what could be the reason for this and why the bands are appearing like this?
Dear Narayana Ruthwek Mamillapalli check your extaction protocol. Did you used a commercial one, or not? It doesn't seem the digestion of DNA, since small bands aren't observed.
Hai Narayana, There is DNA in second gel, it is clearly visible. You can perform PCR with that product. You can also check with nanodrop, if you don't have nanodrop facility, send the DNA in pellet form to any of your friends who have nanodrop.
The DNA bands are not clear because they are degraded, you try to get good results by using careful handling as the degraded results of above image shows the presence of DNA , which protocol you are using and what is the type of DNA is it genomic, cytoplasmic or plastid.
