Phenol and water are immiscible and form two phases a aqueous phase and a phenol phase. Phenol is more dense in which denatured protein partition itself and easily separate when centrifuge.
Phenol extraction is the commonly method used in DNA extraction which remove proteins from Protein-DNA complex. Phenol is immiscible in water. When we vortex phenol and sample, phenol droplets enter into water and denature the proteins. Thus protein can be separated from DNA.
