Why sections shrinks and form lumps during free floating immunofluorescence?

I perform free floating immunofluorescence in mice brain tissue. My sections shrinks and form lumps when I transfer them from TBS to blocking solution or sometimes from primary antibody to TBS. Please provide with the solution. Thanks

Hello Sundeep, it could be the case that your TBS solution has a unsutable concentration. Either you have to dilute it or use this recipe:

6,06 g TRIS HCl (MW 157.6), 1,39 g (MW 121,14), 9 g NaCl in 1 l distilled water, adjust the pH to7,6. Good luck!
