I'm coating liposome with modified chitosan and was expecting the zeta potential of liposome would increase as the concentration of chitosan increase. However, after coating, zeta potential become slightly more towards the negative charge. -30 mV to -35 mV for before and after coating. For preparation method, I just added the chitosan solution dropwise into liposome solution. Anyone facing the same problem? Could suggest me other preparation method?
Why is this a 'problem'? The results are the results if you've conducted the experiment correctly. How have you modified your chitosan? How are you sure that the chitosan has bound to the liposome? Has the ionic balance in the dispersed phase altered before and after the preparation? What was the pH before and after preparation?
Yes, of course why it is a problem, which factors you were considering in your intended use of the preparation? How would that affect!? The question needs little explaining, also like Alan suggested.
Increasing zeta potential in either negative or positive direction shows increasing stability of the particles. So it should not be a problem as suggested by the researchers above.
Is your chitosan modified with a charged, acidic or basic moiety?
Do your liposomes remain liposomes when bound to chitosan (i.e. do substances trapped in the inner volume remain intact)? If it changes to nonliposomal phase upon chitosan binding, then you will get strange readings.
